Overview

Agilent produces a number of cell analysis instruments designed to give life science researchers and drug discovery research scientists better insights into cellular function. The Agilent cell analysis portfolio covers many measurement modalities, including energy metabolism, real-time cell viability, contractility, movement (impedance), and flow cytometry.

Continuously monitor cell response in a 96-well format on the xCELLigence ePlates. Obtain continuous cell assay time points over the course of the entire experiment. Simple label-free protocol measures cell behavior changes continuously from seconds to days. Biosensor-free viewing window facilitates microscopy techniques.

xCELLigence instruments operate in a standard CO2 cell culture incubator and the control unit is housed outside the incubator. User friendly software allows for real-time control and monitoring of the instrument with real-time data analysis functions.

The Agilent xCELLigence E-plate 96 is compatible with the RTCA SP and MP stations. The E-plate VIEW 96 has four center biosensors removed to aid in microscopy. The E-plate 384 well is compatible with the RTCA HT station, and the E-plate Cardio 96 is recommended for use with the RTCA Cardio station.

ACEA offers different types of plates for various purposes.

• E-Plates:
Simple label-free measurements with the usage of a 16-, 48- or 96-well format. These disposable devices can be used for real-time cell analysis with the xCELLigence instruments. The gold biosensors, covering approximately 80% of the well bottom, enable dynamic monitoring.

• E-Plates PET:
Cell analysis can also be performed in plates in which the biosensors are embedded in polyethylene terephthalate (PET) instead of glass. Although cell adhesion to, and proliferation on, PET seems to be similar to glass, some cell types prefer on substrate over the other

• E-Plate inserts:
The 16-well format enables investigation of cell interactions while maintaining the cells in separate compartments. Co-culture of different cell types can be used to measure indirect interactions between cells. The 16-well E-plate insert is compatible with multiple E-plate formats.

• CIM-Plates:
The xCELLigence RTCA DP system makes use of Cell Invasion and Migration plates (CIM-plates). The plate is integrated with a Boyden chamber to provide quantitative kinetic data for cell migration and invasion. The cells pass a membrane when moving towards the chemoattractant in the lower chamber to adhere to the biosensors. This will cause a change in signal which correlates with the number of cells attached.

How is xCELLigence RTCA used for killing assays? The xCELLigence Real-Time Cell Analysis (RTCA) instrument monitors cell killing in real time. Simply add target and immune effector cells to the patented microtiter plates (E-Plates), load the instrument, and start reading. The biosensors embedded in the bottom of each well allow for the automated kinetic measurement of changes in cell number, size, and substrate attachment quality.

Target cell death can be measured continuously and automatically from hours to days without the use of labels.





Simple and high throughput workflow
The xCELLigence Real-Time Cell Analysis (RTCA) instruments utilize patented microplates (E-Plates) containing gold biosensors embedded in the bottom of each well. These biosensors continuously and noninvasively monitor changes in cell number, cell size, and cell-substrate attachment quality.

Cancer Immunotherapy
Immune cell-mediated tumor cell killing can involve the components of both the innate and adaptive immune systems, including natural killer (NK) cells, cytotoxic T cells (MHC-dependent), antibodies secreted by B lymphocytes, engineered antibodies such as bispecific antibodies and bispecific T cell engagers (BiTEs), and genetically engineered T cells targeting specific tumor antigens (e.g., CAR-T, MHC-independent). Cytotoxicity Overview
xCELLigence real time cell analysis (RTCA) uses biosensors on the bottom of E-Plate wells to continuously monitior the number of cells present, their size/morphology, and how tightly they are interacting with the plate surface. Cytotoxic responses nearly always involve biochemical changes that directly, or indirectly, affect one or more of the these parameters. Consequently, xCELLigence is able to monitor cytotoxic responses resulting from an exceptionally wide range of molecular targets (Figure 1).

Cell Barrier Function
The impedance-based xCELLigence Real Time Cell Analysis (RTCA) assay provides a label-free automatic alternative to solute permeability assay and transendothelial electrical resistance (TEER) assay. RTCA assays provides continuous monitoring of cell barrier functions in response to disease state, inflammation, and compound treatment.
